I an not able to understand that, how at time - 5:06 the slope is negative, like (J(w), w) as (-2, 1). Please clarify my doubt ASAP. Thank you

You might consider it this way: if we increase w from that point, then J will decrease, therefore dJ/dw is negative. Since J is decreasing, we have a negative sign in -2.
If, in another point, we increase w, and J will increase, then we say the slope is positive.
Therefore we need to think about how J will react when we increase w.
